AN INVESTIGATION AND EVALUATION STUDY OF ANTIMICROBIAL ACTIVITY FROM COELOMIC FLUID OF INDIAN EARTHWORM PHERETIMA POSTHUMOUS

Abstract:

 

In the current study we have identified and evaluated potent antimicrobial activity from the coelomic fluid of Indian Earthworm Pheretima posthumous. The coelomic fluid of earthworm was extracted in 50mM Phosphate Buffer pH 8.5. The coelomic fluid was further characterized as serine protease by casein hydrolytic agar plate assay. This protease had shown enormous antimicrobial activity which was quite stable in various pHs, temperatures and in the presence of different inhibitors. This protease has shown maximum activity at 40oC which was stable up to the 70oC and pH of 8.0 which further ranges 5.0 -10.0. The protease was quite stable and active for long storage in lower temperature. We have also evaluated antifungal activity in various conditions. Inhibitors and chelating agents, Aprotinin, Sodium azide and EDTA respectively have shown very nominal effect on the stability and activity of coelomic fluid. We have evaluated the antimicrobial activity in different bacterial strains including psychrophilic, mesophilic thermophilic bacteria. The coelomic fluid has shown satisfactory antifungal activity in the different condition of pH and temperatures. Presently our team is working on the purification of crude coelomic fluid to determine molecular weight and other biophysical characteristic of antimicrobial compound form earthworm.
